Reporter sequence of delay system
BBa_K3221204 is consisted of pMET3(BBa_K3221201), GAL4(BBa_K3221208) and pGAL1-yeGFP(BBa_K3221200). In our project, MET3 promoter and GAL4-pGAL1 construct the delay system to ensure the adequate cultivation of yeast before the expression of cns1-cns2 fusion protein and yeGFP is used as the reporter. And the delay efficiency is adjustable and can be controlled by the presence of different concentrations of methionine in the medium. This system can be also used for other delay systems in Saccharomyces cerevisiae.
